Use extreme caution moving the X-Y stage if the sample holder has a lip on it that extends above the sample surface like a petri dish. The SPM head can be destroyed if the sample stage is moved such that the tip holder crashes sideways into the lip of your sample holder. Also, check that the position of the lip of the sample holder will not interfere with the movement of the Z-stage.

Introduction The BioScope is an exciting new tool for the investigation of biological surfaces. The optical microscope allows quick location of regions of interest biozcope a sample which can then be imaged at higher resolution with the scanning probe microscope.
The microscope SPM head assembly may also be detached and operated separately from the inverted microscope for larger samples see Figure 3.
The SPM creates high-resolution images of a sample surface by scanning a sharp tip manul the sample. Images mxnual the sample surface are created by raster-scanning the cantilever and tip across the surface while monitoring variations in the cantilever motion. The results of the cantilever measurements are recorded and can be displayed as false color images or as threedimensional plots. The BioScope system consists of a rigid metal frame that suspends the Dimension SPM head above the inverted optical microscope.
The SPM head may be moved towards the sample or retracted from the sample with an automated and motorized translation stage.
The BioScope is provided with metal sample holders for glass slides and petri dishes. These sample holders mount under the SPM head and can be translated with a manual X-Y stage, located on the right side of the BioScope.
Introduction Normally the user positions the sample until a region of interest is located under the SPM tip, then the SPM is engaged onto the region of interest. With optional cantilever tips and tipholders, additional capabilities such as magnetic force microscopy MFMelectric force microscopy EFMand force modulation are available.
Microscope Head The Dimension SPM head provides accurate imaging of a stationary sample while scanning the integrated detector probe assembly above the sample. The Dimension head is very effective in imaging large samples too large or heavy to scan by movement of the sample. Patents covering the technology developed in this head are pending.

The integrated scanner head consists of the following subassemblies: Laser Diode Stage— A kinematic tilt stage is used to position the laser beam onto the cantilever. It consists of the laser diode, collimator, focusing lens, baseplate, and the Biosocpe and Y laser diode positioners.
The X positioner knob moves the beam parallel to the major axis of the cantilever substrate. The Y positioner knob moves the beam perpendicular to the major axis of the cantilever substrate. The use of these positioners is described in Chapter 5.
